Four test tubes were labeled “H2O A”, “H2O B”, “Enz A”, and “Enz B”. In the test tubes labeled “Enz A” and “Enz B” 1 mL of catalase was pipetted into each, and in the test tubes labeled “H2O A” and “H2O B” 6mL of deionized water was pipetted into each. Next the two test tubes labeled A were placed into an ice bucket, and the test tubes labeled B were placed in a 50°C water bath. These were left in their respected locations for 5 minutes each, and then the temperature of the test tubes in the ice bucket were recorded. Next the “Enz A” test tube was mixed with the “H2O A” test tube and 100µL of guaiacol and 150µL of .1% H2O2 were added to this mixture using a micropipette. The test tube was then covered with aluminum foil and inverted to mix the solution and then placed in a curvette and into the spectrometer. The initial absorbance was recorded and the curvette was placed back in the ice bucket. Then the contents of the test tubes labeled B were mixed and 100µL of guaiacol and 150µL of .1% H2O2 were added to this mixture using a micropipette. This was then placed in a curvette and into the spectrometer and the initial absorbance was recorded and the curvette placed back in the water bath. Apparatus: Spectrophotometer (UV-1201), cuvettes, water bath (set at 37°C), 200µl and 1000µl micropipettes and test tube
By removing one milliliter of the stock solution to the first test tube, and repeating the same process for the other two tubes allowed us compare concentrations and know what the dilution factor would be when placed into the dialysis tubing. All three tubes had the same amount of water (4mL of H2O) added. Once the solution for each test tube was mixed well, we transferred 2 mL of each concentration into the dialysis bag. We secured each piece (3 in total) of the dialysis tubing to ensure that once the concentration was added that it would not leak out of either end. Three stir plates were used, all set at the same speed, and the dialysis bags filled with different concentrations were added. Before we began to measure the absorbance rate, we did take a sample of the water immediately after placing the dialysis tubing into each beaker. We observed each beaker over a period of 25 minutes. At each five-minute interval, we removed one milliliter of the solution and placed it into a cuvette. The cuvette sample was then placed in the spectrophotometer to calculate the absorbance rate of each sample. After each sample was measured, all data was recorded in the lab manual. Once all measurements had been recorded we used the absorbance rate to help calculate the concentration. The function of the gel is so that the DNA strands can separate out. The electrophoresis process is necessary because, due to DNA having a negative charge naturally it will be attracted to substances that are positively charged, so by positioning the gel, with the DNA samples injected into its wells, facing towards the positive end of the electrophoresis apparatus with the DNA samples on the opposite end closest to the negative end, the DNA strands with gradually move down the gel and separate based on their band size. Before separating the DNA strands can be accomplished, the gel must first be made accordingly as well as the samples. In order to make the gel, agarose powder and a buffer must be mixed and cooked together first. The buffer that was used in this experiment is the 1X TAE buffer.
